چکیده

Definition Peptide nucleic acid (PNA) is an artificial mimic of nucleic acid, which was invented more than a decade ago (7). The molecule combines many features that are similar to those of natural nucleic acids with characteristics that are rather different or even unique to PNA. First and foremost, N-(2-aminoethyl)-glycine units linked by ▶amide bonds substitute the (deoxy)ribosephosphate backbone and the nucleobases are attached via a methylene carbonyl link to the glycine moiety. Nevertheless, the distances and angles within PNA oligomers much resemble the conformation of nucleic acids (Fig. 1). One format of application is the microarrays that are made for a highly parallel analysis of nucleic acids. To this end, very many different PNA molecules are attached onto a small planar surface, e.g. a microscope slide. Upon addition of a DNA or RNA sample, hybridisation occurs on the chip at PNAoligomers that are of complementary sequence to molecules in the sample. Recorded by detecting a label that is attached to the DNA or RNA, various forms of information can be deduced from such studies, such as for example, sequence variations or changes in the amount of an individual RNA molecule within a complex mixture.

Development of PNA microarrays for gene diagnostics with TOF-SIMS

We have used time-of-flight secondary ion mass spectrometry (TOF-SIMS) to examine different immobilization processes of peptide nucleic acid (PNA) and their influence on the hybridization efficiency of unlabeled DNA fragments to complementary PNA. Two different approaches have been used to immobilize PNA onto Au surfaces. One method was to immobilize thiolated PNA in a single step reaction to t...

Impedance-based detection of DNA sequences using a silicon transducer with PNA as the probe layer.

Electrochemical impedance measurements were used for the detection of single-strand DNA sequences using a peptide nucleic acid (PNA) probe layer immobilized onto Si/SiO2 chips. An epoxysilane layer is first immobilized onto the Si/SiO2 surface. The immobilization procedure consists of an epoxide/amine coupling reaction between the amino group of the PNA linker and the epoxide group of the silan...

Mass spectrometric characterization of DNA microarrays as a function of primary ion species

Recent studies have shown TOF-SIMS to be an appropriate method for the detailed examination of the immobilization process of PNA and its ability to hybridize to unlabeled complementary DNA fragments. Unlabeled single-stranded DNA was hybridized to Si wafer biosensor chips containing both complementary and non-complementary immobilized PNA sequences. The hybridization of complementary DNA could ...

Isolation and characterization of mitochondria-rich cell types from the gill of freshwater rainbow trout.

A magnetic cell separation technique (MACS) was developed for isolating and characterizing peanut lectin agglutinin positive (PNA(+)) cells from rainbow trout gills. Percoll density separated mitochondria-rich (MR) cells were serially labeled with PNA-FITC and an anti-FITC antibody covalently coupled to a 50-nm iron particle and then applied to a magnetic column. PNA(+) MR cells were enriched t...

The first crystal structures of RNA–PNA duplexes and a PNA-PNA duplex containing mismatches—toward anti-sense therapy against TREDs

PNA is a promising molecule for antisense therapy of trinucleotide repeat disorders. We present the first crystal structures of RNA-PNA duplexes. They contain CUG repeats, relevant to myotonic dystrophy type I, and CAG repeats associated with poly-glutamine diseases. We also report the first PNA-PNA duplex containing mismatches. A comparison of the PNA homoduplex and the PNA-RNA heteroduplexes ...
